Localization of Thioredoxin-Interacting Protein in Aging and Alzheimer’s Disease Brains

چکیده

Thioredoxin-Interacting Protein (TXNIP) has been shown to have significant pathogenic roles in many human diseases, particularly those associated with diabetes and hyperglycemia. Its main mode of action is sequester thioredoxins, resulting enhanced oxidative stress. The aim this study was identify if cellular expression TXNIP aged Alzheimer’s disease (AD) brains correlated pathological structures. This employed fixed tissue sections protein extracts temporal cortex from AD control brains. Studies light fluorescent immunohistochemical techniques using the monoclonal antibody JY2 Immunoblots were used quantify relative amounts brain extracts. major finding identification immunoreactivity selective neuronal populations structures, non-AD In brains, less but increased numbers TXNIP-positive plaque-associated microglia observed. Immunoblot analyses showed no increase levels samples tested. conclusion, identified altered patterns progression pathology.

Thioredoxin-interacting protein suppresses bladder carcinogenesis.

Thioredoxin-interacting protein (TXNIP), which has a tumor-suppressive function, is underexpressed in some human cancers. The function of TXNIP in vivo in carcinogenesis is not fully understood. Here, we show TXNIP to be downregulated in human bladder cancer according to grade and stage and also that loss of TXNIP expression facilitates bladder carcinogenesis using a mouse bladder cancer model....

Nitrosative/Oxidative Stress Conditions Regulate Thioredoxin-Interacting Protein (TXNIP) Expression and Thioredoxin-1 (TRX-1) Nuclear Localization

Thioredoxin (TRX-1) is a multifunctional protein that controls the redox status of other proteins. TRX-1 can be found in the extracellular milieu, cytoplasm and nucleus, and it has distinct functions in each environment. Previously, we studied the intracellular localization of TRX-1 and its relationship with the activation of the p21Ras-ERK1/2 MAP Kinases signaling pathway. In situations where ...

Thioredoxin Interacting Protein (TXNIP) and Pathogenesis of Diabetic Retinopathy.

Chronic hyperglycemia (HG)-associated reactive oxygen/nitrogen species (ROS/RNS) stress and low grade inflammation are considered to play critical roles in the development of diabetic retinopathy (DR). Excess glucose metabolic flux through the aldose reductase/polyol pathway, advanced glycation end product (AGE) formation, elevated hexosamine biosynthesis pathway (HBP), diacyl glycerol/PKC acti...
